《赖汉的幸福指数》剧情主要讲述的是赖汉“赖亚洲”逐渐成长为一个肯吃苦、肯奋斗，有担当的男子汉的故事，同时剧中还穿插很多搞笑、幽默的桥段，呈现出了一部现实喜剧。
当兵十年的柏胜兵在退伍回到家乡南湾村后，发现家乡在他父亲柏保国的领导下，与十年前几乎毫无变化。他放弃了城里武装部的工作，在自愿下乡支教的城里女大学生柯盛楠帮助下，成功当上了代理村长和村支部书记。钱砚开因没能被选上村长而记恨柏胜兵。大学生才文秀，一心追求柯盛楠，吹嘘自己是城里某公司茶园项目经理而引发一系列故事。在合力帮助村子发展的过程中，柯盛楠渐渐喜欢上了柏胜兵，钱砚开的儿子钱串子也喜欢柯盛楠。才文秀提出与柏胜兵公平竞争。在“万人大采茶”民俗旅游节上，当导游的钱串子与外国姑娘一见钟情。才文秀在城里的茶园项目公司工作，与董事长的女儿苏慧相识相恋。村民集体搬迁的第一天，柏胜兵与柯盛楠、才文秀与苏慧、钱串子与莉莉、钱砚开与王春兰举行了盛大婚礼，南湾村从此掀开了崭新的一页。
